ReaPan S34 for the Quantitation of CD34+ Cells

The ReaPan S34 assay is intended for the determination of the concentration of CD34+ cells in cord blood, bone marrow samples, and apheresis products using flow cytometry. The ReaPan CD34 reagent is formulated in buffered saline with 0.1% sodium azide and stabilizers. It contains fluorescently labeled CD45 and CD34, as well as 7-aminoactinomycin D. A precise number of fluorescent counting beads are included in the ReaPan CD34 reagent to allow single-platform determination of absolute CD34+ stem cell counts.

The ReaPan S34 reagent is provided in dried form dispensed in flow cytometer compatible sample tubes. There is enough reagent in each tube to process one sample.


CD34 is a surface glycoprotein expressed on hematopoietic stem and progenitor cells. Cancer patients, following ablative therapy, receive infusions of CD34+ cells to reestablish their hematopoietic system. The number of CD34+ cells infused correlates with the time required to reestablish the hematopoietic system. Enumeration of CD34+ cells in peripheral blood may be used to determine if mobilization of CD34+ cells has occurred. Enumeration of CD34+ cells in apheresis product may also be used to determine if a sufficient number of CD34+ cells have been collected for successful engraftment.

 The analysis is based on ISHAGE/ISCT protocols for HPC enumeration by flow cytometer, taking into consideration the following guidelines:

  • HPC's are CD34+ (bright) and CD45+ (dim)
  • HPC's exhibit low side scatter (SSc) and moderate to high forward scatter (FSc)
  • HPC's exhibit morphological features similar to that of lymphocytes
  • CD34+ HPC's enumeration is based on an absolute bead count using a precise number of fluorescent beads.
